Effective and validated animal models are valuable to investigate the pathogenesis and potential therapeutics for human diseases. There is much concern for diabetic retinopathy (DR) in that it affects substantial number of working population all around the world, resulting in visual deterioration and social deprivation. In this review, we discuss animal models of DR based on different species of animals from zebrafish to monkeys and prerequisites for animal models. Despite criticisms on imprudent use of laboratory animals, we hope that animal models of DR will be appropriately utilized to deepen our understanding on the pathogenesis of DR and to support our struggle to find novel therapeutics against catastrophic visual loss from DR. 相似文献

A process for high level production of rapamycin by Streptomyces hygroscopicus using statistical designs and feeding strategy was developed. The amino acids (i.e. Lys, Tyr, and Gln) for precursor supply were screened out in the initial phase of fermentation. The optimum levels determined with Box-Behnken design were Lys 20, Tyr 4, and Gln 3 g/l. In the rapamycin biosynthesis phase, the important component, ammonium sulphate, was also identified. A novel two-stage feeding strategy was developed successfully to increase the flux of rapamycin biosynthesis, in which the optimized amino acid components were fed in the initial phase of fermentation, and then switched to feed 2 g/l ammonium sulphate at 72 h. The maximal rapamycin production reached 860.6 mg/l in a 7 l fermentor, which was 182 % higher than that of the control. This was the first report to integrate precursor engineering and cell physiological regulation methods to optimize rapamycin production. 相似文献

Brown cotton is a kind of naturally colored cotton. Because of less processing and little dying, it is more friendlier to
environment than white cotton. For brown cotton, pigment accumulation in fiber is one of the most important characteristics.
In this study, we selected a brown fiber line and a white fiber cultivar to determine the factor that affects the pigmentation
in brown fiber. Accordingly, fibers were collected to verify the presence of PAs by p-dimethylaminocinnamaldehyde (DMACA) and toluidine blue O (TBO) staining. The PAs content and related genes expressions were
determined. As a result, there were obvious differences on the aspect of PAs synthesis in fiber between white cotton and brown
cotton. For white fiber, the PAs content reached maximum at 5 DPA, and then gradually decreased to zero. But for brown fiber,
the PAs content was increased from 5 to 15 DPA stage, and reached the maximum at the 15 DPA stage, then gradually decreased
from 15 to 40 DPA stage. On the contrary, in white cotton, PAs were synthesized in the whole developmental stage from 5 to
40 DPA. And PAs content in brown fiber were far more than that in white fiber, which may be the reason why the brown pigment

A peptide ranging from residues 229 to 240 (ENESANEGYRFG) of D1 protein was synthesized by stepwise solid-phase method. Resolution enhancement techniques were combined with band curve-fitting procedures to quantitate the FTIR spectra in the amide I' region (1700-1600 cm-1). FTIR analysis showed that DCMU induced drastic structural modification with a relative decrease of the unordered structure and turns, and a substantial increase of α-helix, which indicated that a much more compact structure was formed when DCMU was applied. To study the structure-activity relationship between pentanoic acid analogues and the inhibition of fatty acid oxidation, a number of 4-pentenoic and methylenecyclopropaneacetic acid derivatives were prepared. All compounds inhibited palmitoylcarnitine oxidation in rat liver mitochondria, with 50% inhibition occurring at a concentration between 6 and 100 microM. However, only methylenecyclopropaneacetic acid (MCPA) and spiropentaneacetic acid (SPA) showed in vivo inhibitory activity in rats as indicated by the occurrence of dicarboxylic aciduria. Rats treated with SPA excreted metabolites derived only from fatty acid oxidation whereas MCPA-treated rats also excreted metabolites derived from branch-chained amino acid and lysine metabolism. SPA is a specific inhibitor of fatty acid oxidation without affecting amino acid metabolism. The site of inhibition is medium-chain acyl-CoA dehydrogenase (MCAD). In contrast, MCPA inhibited both MCAD and short-chain acyl-CoA dehydrogenase with a stronger inhibition toward the latter. The inhibition of fatty acid oxidation by both inhibitors was partially reversible by glycine or l-carnitine. Since SPA does not form a ring-opened nucleophile such as that proposed for MCPA in the inhibition of FAD prosthetic group in acyl-CoA dehydrogenases, we propose that the irreversible inhibition by SPA occurs by a tight complex without forming a covalent bond to the isoalloxazine ring in FAD. 相似文献

The pollination system of Holcoglossum rupestre was studied in northwestern Yunnan. The species is self-compatible but pollinator-dependent and offers nectar as a reward to visitors. A species of beetle, Hybovalgus bioculatus Kolbe, is identified as the pollinator among all visitors observed. The beetles carried the pollinia in two different positions, i.e. on the buttock and the foreleg, and pollinated the flowers by two different mechanisms. The analysis suggests that Holcoglossum rupestre and its pollinator may be in an unstable pollination relationship. The natural fruit set is low, which is attributed to the low visiting frequency of pollinator and the effective mechanism to prevent self-pollination. 相似文献
